The numbers represent which carbon the substituents are added to. You number the carbons on the longest chain and it could start on the right or left side, but in the end, you number it so that the substituent is bonded to the lowest number carbon.

MnO4 is flipped because it is the half-reduction that is being oxidized. MnO4 is loosing electrons during the reaction, making it the anode and the anode is always negative.

The cell potential describes how much work can be produced from the energy converted from the system. It is compared to the standard hydrogen electrode because this electrode serves as a reference point when calculating/comparing data.

Because the glass of water is loosing 100J of energy and the change in S = Qrev/T, if Q decreases, then so does S because they're proportional.
