Can a rate be negative and why?

Rate cannot be negative. It is defined as positive. Similarly, we usually consider speed as positive.

I'm pretty sure that the rate of a chemical reaction can only be positive.

No a rate cannot be negative. A reaction cannot progress at -2 M/s.
