Because of the varying ways to calculate delta of frequency which is the most optimal in doing so? and should Rydberg's constant be positive or negative when doing so?

It all depends on the situation and the type of problems they ask. All of the equations that help calculate contain drastically different variables and information that is needed to solve it. Therefore, it really depends on the information given. Also, Rydberg's constant is a positive value when calculating frequency in rydbergs equation.
